Substrate anchoring and the catalytic power of enzymes.
نویسنده
چکیده
Evidence from published nuclear magnetic resonance studies of enzyme-substrate and enzyme-inhibitor systems systems shows that substrates are confined at the active site of the enzyme, have a relatively long residence time, and tumble in solution as an enzyme-substrate complex. The consequences of this "substrate anchoring" with regard to the catalytic power of enzymes are considered. It appears that as a result of the relatively slow motions, the probability of existence of the "activated complex" in the enzymatic reaction may be increased and thus the reaction rate may be accelerated by factors of between 10(6) and 10(9). It is estimated that the combined effect of substrate anchoring, desolvation, and charge compensation may lead to reaction-rate accelerations by factors of between 10(9) and 10(8).

Organophosphate compounds and their biodegradation; using enzymes as an increased efficiency approach
Organophosphorus compounds are widely used in pesticides, insecticides in agriculture and as nervous chemical agents. These chemicals inhibit the acetylcholinesterase enzyme activity that is responsible for the nervous impulse in organisms. This effect leads to an increase in acetylcholine level and finally neuronal complications. Many methods are used to degrade and decontaminate these compou...
متن کاملThe Role of Highly Conserved Tryptophan in the Sixth Conserved Region at Substrate Specificity of α- amylase
Early in this study, an α-Amylase from Bacillus megaterium WHO (BMW) was isolated from hot springs of Ramsar (North of Iran), and its gene was cloned in E.coli. Based on its conserved sequence regions and substrate specificity, it was classified as intermediary group enzymes with the specificity of oligo-1,6-glucosidase and neopullulanase subfamilies. In the sixth conserved re...
متن کاملBio-inspired Construction of Advanced Fuel Cell Cathode with Pt Anchored in Ordered Hybrid Polymer Matrix
The significant use of platinum for catalyzing the cathodic oxygen reduction reactions (ORRs) has hampered the widespread use of polymer electrolyte membrane fuel cells (PEMFCs). The construction of well-defined electrode architecture in nanoscale with enhanced utilization and catalytic performance of Pt might be a promising approach to address such barrier. Inspired by the highly efficient cat...
متن کاملOrigins of catalysis by computationally designed retroaldolase enzymes.
We have investigated recently reported computationally designed retroaldolase enzymes with the goal of understanding the extent and the origins of their catalytic power. Direct comparison of the designed enzymes to primary amine catalysts in solution revealed a rate acceleration of 10(5)-fold for the most active of the designed retroaldolases. Through pH-rate studies of the designed retroaldola...
متن کاملMolecular basis of enzyme catalysis and control.
Regulatory processes in living systems can be shown to be caused by ligand induced changes in the shape of a protein. By dividing the protein shape changes into molecular parameters a quantitative analysis of the regulation process can be obtained which is shown to correlate with actual data in living systems. The complexity of the analysis increases as the number of subunits in the enzyme and ...
